Ashburn (Sandra Bullock) é uma agente especial do FBI extremamente competente, apesar de ser mal vista pelos colegas de trabalho por ser arrogante e antipática. De olho em uma promoção no trabalho, ela pede ao seu chefe (Demian Bichir) que a encarregue da investigação de um poderoso traficante de drogas em Boston, cuja identidade é desconhecida. Entretanto, logo ao chegar Ashburn decide interrogar um pequeno traficante preso por Mullins (Melissa McCarthy), uma desbocada policial local que não aceita ordens de ninguém. Não demora muito para que as duas batam de frente, mas elas precisam encontrar um meio de trabalhar juntas.
